FV3000 confocal laser scanning microscope (Evident). The system is equipped with 4 excitation lasers lines: 405, 488, 561 and 640 nm (Coherent), a galvanometric scanning head and 4 spectral detectors, allowing to perform simultaneous imaging of up to 4 fluorescence channels. Brightfield images can be acquired using a scanning laser line and a transmitted light detector. The IX 83 inverted microscope body (Olympus) includes a motorised sample stage for multi-area mosaic image acquisition.
The objective lenses used were the following:
– UPLXAPO 4x/0.16 NA (Olympus)
– UPLXAPO 10x/0.4 NA (Olympus)
– UPLXAPO 20x/0.8 NA (Olympus)
– A silicone oil immersion UPLSAPOXS 30x/1.05 NA (Olympus)
– An oil immersion lens UPLXAPOO 60x/1.42 NA (Olympus).
